If you run "Audio Midi Setup" (search for this using Spotlight) this gives a list of all your sound inputs and outputs.
Is there a icon of a loud-speaker next to "Built-in Output" (or whatever sound-output you want to use)? If not, right click on "Built-in Output" and choose "Use this device for sound output".
Have you ever been using an optical cable from the output jack? Sometimes this doesn't quite connect properly, and the Mac doesn't recognise it inserted/not-inserted. Try inserting/removing a headphone jack a few times.
Are you using loudspeakers built into an external screen? Particularly the large 27 inch Apple cinema display had a bug where the audio would stop working over USB connection. There is a firmware upgrade to help correct this.
And check you haven't turned-off the sound accidentally - press the volume up key.

I've seen that before but it was a long time ago - back in 10.5 (leopard) days. So some questions;
- which version of Mac OSX did you upgrade to?- what exact model of Macbook is it? Most interestingly, does it have a battery that you can easily remove?- does it still make the basic Mac "gong" sound early in the process of turning it on?- have you tried plugging in some headphones to see if there is sound at the headphone jack?

Would suggest you take it to a Mac store, the experts there will probably know what to do, and their advice is free! There is one at Cap3000.
